Antitumor activity of sugar-modified cytosine nucleosides.
Nucleoside analogues which show antimetabolic activity in cells have been successfully used in the treatment of various tumors. Nucleosides such as 1-beta-D-arabinofuranosylcytosine (araC), 6-mercaptopurine, fludarabine and cladribine play an important role in the treatment of leukemias, while gemcitabine, 5-fluorouracil and its prodrugs are used extensively in the treatment of many types of so...
متن کاملSynthesis and antiretroviral activity of phospholipid analogs of azidothymidine and other antiviral nucleosides.
Treatment of acquired immunodeficiency syndrome with azidothymidine (AZT, zidovudine) reduces p24 antigenemia, increases CD4 lymphocyte counts, reduces the frequency and severity of opportunistic infections and prolongs life. However, AZT and other dideoxynucleosides do not diminish the ability to isolate human immunodeficiency virus (HIV) from peripheral blood mononuclear cells. Failure to cle...
متن کاملSynthesis and antiviral properties of spirocyclic [1,2,3]-triazolooxazine nucleosides.
An efficient synthesis of spirocyclic triazolooxazine nucleosides is described. This was achieved by the conversion of β-D-psicofuranose to the corresponding azido-derivative, followed by alkylation of the primary alcohol with a range of propargyl bromides, obtained by Sonogashira chemistry. The products of these reactions underwent 1,3-dipolar addition smoothly to generate the protected spiroc...
متن کاملSynthesis and antiviral activity of a series of new cyclohexenyl nucleosides.
A series of new cyclohexenyl nucleosides is synthesized by coupling the heterocyclic bases with a protected cyclohexenyl precursor under Mitsunobu conditions. The compounds were evaluated for their antiviral and cytostatic activity. Pronounced activity against herpes simplex virus type 1 and type 2 was observed for the 2,6-diaminopurine analogue.
متن کاملEnantiomeric Synthesis of Novel Apiosyl Nucleosides as Potential Antiviral Agents
A series of 2',3'-dideoxy-3'-fluoro-D-apiosyl nucleosides 15, 16, 17 and 18 were synthesized enantiomerically with L-Gulonic-γ-lactone as the starting material. The reduction of butenolide 1 with DIBAL-H followed by the Luche procedure afforded the allylic alcohol 2. Ozonolysis and the reduction of compound 4 induced the cyclized lactol, which was acetylated to give the acetate 7. Condensation ...
